Molecular Pathways: Mechanism of Action for Talimogene …

3/1/2016  · Mechanism of action for T-VEC. T-VEC preferentially replicates in cancer cells because of disrupted PKR activity ( 1 ), which may be lower due to overactive Ras signaling. PKR signaling regulates cell proliferation by phosphorylating eIF-2, which prevents delivery of Met-tRNAi to ribosomes blocking protein translation.

T-VEC Mechanisms of Action and Preclinical Activity Oncolytic viruses include wild-type and modified live viruses and represent an innovative approach to cancer immunotherapy.

T-VEC, entering the cancer cells through the herpes virus glycoproteins on the cell surface, favorably replicates and ultimately triggers cell lysis [32]. In this way, tumor-derived anti-gens (TDA), GM-CSF and new viral particles are released allowing just one-time infection of surrounding tumoral cells and enhancing TDA spreading [33].

This review highlights the dual mechanism of action for talimogene laherparepvec (direct tumor cell lysis and stimulation of local response in tumor microenvironment and systemic immune response …
